Variability of Red Clover Genes for Nitrogen Fixation and its Use by Molecular Selection

The symbiosis between plants and rhizobia is controlled by key genes involved in the set up and regulation of nodulation and biological nitrogen fixation (BNF). Intraspecific variation in nitrogen fixation is determined by the genotypic profile at loci with key genes of nitrogen fixation. Recent studies have identified candidate allelic variants at selected loci associated with fixation efficiency. Before applying the associated SNPs to breeding through marker-assisted selection, it is advisable to verify the predictive power of the associated SNPs in a broad parental and selected progeny populations. In this study, we thus validated 8 previously proposed SNP markers associated with nitrogen fixation efficiency in two large populations. Only for the SNP marker that lies in the gene for the beta tubulin on chromosome 3 predominantly expressed in nodule we confirmed a clear association across generations. This SNP marker is suitable for application of marker-assisted selection for fixation efficiency as it explains part of the variability in nitrogen fixation levels. Knowledge of the genetic variability within BNF candidate genes and the verification of candidate associated SNPs will benefit molecular selection and breeding of red clover.
