The herbicides inhibiting the photosystem II (PSII) still represent a basic tool of weed control. Their overuse brings serious environmental and health risks. Since chromatographic methods and immunoassays are not suitable for prescreening assay, we haverecently designed a biosensor based on isolated PSII from thermophilic cyanobacteria Synechococcus elongatus1. The performance of this set-up is restricted by crosslinking the PSII in a gel matrix of bovine serum albumine (BSA) and glutaraldehyde (GA).Gel entrapment resulted in slow mediator diffusion and decreased sensitivity to herbicides. An oriented monolayer of PSII could bring entirely new properties such as fast mediator transport, increased sensitivity and spatially controlled immobilization.Here we present the results of two methods of monolayer immobilization of PSII and prospects for its use as a biosensor for detection of herbicides: (i) Immobilisation based on the recombinant (His)6-tagged PSII;(ii) Immobilisation based
