Pea is an important food and feed and is among the most environmentally friendly crops due to the fixing of nitrogen. Pea breeding has a long tradition in the Czech Republic. The aim of the Czech breeding program is to obtain new genetic materials with emphasis on water and pathogen resistances. In connection with these requirements, the natural properties of the wild species (P. fulvum, P. elatius, P. abyssinicum), including Vavilovia formosa and pea breeding lines, can be used. In vitro multishoot cultures (MSCs) of pea wild type obtained from nodal segments and apex of germinated seedlings were introduced to design bioassays to detect abiotic resistance, eg. drought and pathogenes resistences. The potential for introducing cultures, rooting multiplied shoots and transferring them to ex vitro conditions (micropropagation potential) has been identified. To increase genetic variability of pea, in vitro induced polyploidization with the use of antimitotic agents, interspecific hybridization using protoplast cultures are also used. In the induction of chromosome doubling, we focused on the optimization of the oryzaline concentration and duration of action in Vavilovia formosa. The occurrence of morphological deviations associated with ploidy was recoreded. Protoplast cultures were optimized for cultivation of donor plants (composition of culture medium, plant age) and combinations and concentrations of enzymes inducing cell wall degradation. The work is in progress. The project is the first year of the solution.
