20-hydroxyecdysone prevents oxidative stress damage in adult Pyrrhocoris apterus

  • Original language description

Injections of paraquat into adult P. apterus caused a significant elevation of lipid peroxidation and protein carbonylation and a decline of membrane fluidity in the microsomal brain fraction. Another manifestation of oxidative stress was a depletion ofthe reduced glutathione pool and reduction of the ?-glutamyl transpeptidase activity in the brain extracts. The damaging action of paraquat on the brain was counteracted by simultaneous injection of 20-hydroxyecdysone (20E). 20E restrained lipid peroxidation and the formation of protein carbonyls, ameliorated changes in microsomal membrane fluidity, enhanced the level of reduced glutathione, and upregulated the activity of ?-glutamyl transpeptidase. At the organismic level, 20E curtailed three detrimental effects caused by paraquat injection: the disappearance of a blood protein, the suppression of fecundity and egg hatchability, and the shortening of adult life span.

  • Czech name

20-hydroxyekdyson zabraňuje poškození organismu oxidačním stresem u dospělců Pyrrhocoris apterus

  • Czech description

Injekce paraquatu do těla dospělce P. apterus způsobila signifikantní zvýšení lipidové peroxidace a proteinové karbonylace, a dále pokles fluidity membrán v mozku. Oxidační stress organismu byl dále manifestován spotřebováním zásob redukovaného glutathionu a snížením aktivity ?-glutamyl transpeptidasy v mozkových extraktech. Popsaný vliv paraquatu byl eliminován injekcí 20-hydroxyekdysone (20-E), který zabraňoval peroxidaci lipidů a tvorbě protein karbonylů, zlepšoval fluiditu membrán, zvyšoval hladinuredukovaného glutathionu i aktivitu ?-glutamyl transpeptidasy. Na úrovni organismu 20E snižoval trojí účinek injekce paraquatu: pokles hladiny proteinů hemolymfy, snížení plodnosti a líhnivosti vajíček a zkrácení délky života dospělců.
