    The 2?-O-ribose methyltransferase for cap 1 of spliced leader RNA and U1 small nuclear RNA in Trypanosoma brucei

    The function of cap 1 2'-O-ribose methylation is not known in any eukaryotic system, although it is a conserved modification. In kinetoplastid flagellates trans-splicing of spliced leader (SL) to polycistronic precursors conveys the hypermethylated cap 4to all nuclear messenger RNAs. We report the first eukaryotic cap 1 2'-O-ribose methyltransferase, TbMTr1, a member of a conserved family of viral and eukaryotic enzymes. Recombinant TbMTr1 methylates the ribose of the first nucleotide of a m7G-capped substrate. Knockdowns and knockouts of TbMTr1 in Trypanosoma brucei grow normally, while displaying loss of 2'-O-ribose methylation at cap 1 on SL and U1 snRNA. TbMTr1 knockouts have an accumulation of cap 0 substrate, however spliced messenger RNA is modified at position 4 and lacks 2'-O-ribose methylation at position 1. Accumulation of 3'-extended SL indicates a delay in processing, and suggests a synergistic role for cap 1 in maturation of substrate SL RNA.

    Dosud není známa funkce 2?-O-ribozové metylace v čepičce typu 1 u žádného eukaryota, ačkoli se jedná o vysoce konzervovanou medifikaci. Díky sestřihu formou trans z polycistronických prekurzorů prostřednictvím spliced leader (SL) dochází u kinetoplastidových bičíkovců k přenosu hypermetylované čepičky typu 4 na všechny jaderné poslíčkové RNA. V této práci jsme charakterizovali jako první eukaryotickou čepičkovou 2?-O-ribozovou metyltransferázu TbMtr1, která je členem konzervované rodiny virových a eukaryotických enzymů. Knockdownové a knockoutové linie Trypanosoma brucei pro gen TbMTr1 rostou normálně, ačkoli se u nich nachází ztráta 2?-O-ribozové metylace na čepičce typu 1 u SL a U1 malých jaderných RNA. U knockoutové linie pro TbMTr1 došlo k akumulaci substrátu čepičky O, ačkoli je sestřihaná poslíčková RNA modifikována v pozici 4 a postrádá metylaci 2?-O-ribozy v pozici 1.
