Atf3 links loss of epithelial polarity to defects in cell differentiation and cytoarchitecture

  • Original language description

Apico-basal polarity is critical for integrity and function of epithelial tissues and its loss is one of the hallmarks of cancer. This study shows that in the fruit fly Drosophila melanogaster breakdown of epithelial polarity engages Activating transcription factor 3 (Atf3), a protein that directly binds the DNA and regulates gene expression. As such, the abnormally induced activity of Atf3 drives many defects in the epithelial function and differentiation, whereas removal of Atf3 in this context is beneficial. Given the evolutionary conservation of Atf3 between flies and humans, these findings may contribute to a better understanding of diseases stemming from compromised epithelial polarity.
