    We have developed a simple method to resolve the sex chromosome constitution in females of Lepidoptera by using a combination of genomic in situ hybridization (GISH) and fluorescence in situ hybridization with (TTAGG)n telomeric probe (telomere-FISH). Inpachytene sex chromosomes, GISH differentiated W heterochromatin and telomere-FISH detected the chromosome ends. With this method we showed that Antheraea yamamai has a W-Z sex chromosome pair. In Orgyia antiqua, we confirmed the presence of neo-W and neo-Z chromosomes. Orgyia thyellina females displayed a neo-ZW1W2 sex chromosome constitution. A neo-WZ1Z2 trivalent was found in females of Samia cynthia subsp. indet. Whereas S. c. walkeri showed a neo-W/neo-Z bivalent and S. c. ricini showed a Z univalent (a Z/ZZ system).     Rozlišení konstituce pohlavních chromosomů pomocí GISH a telomerické FISH u některých druhů motýlů

    Vyvinuli jsme jednoduchou metodu pro rozlišení konstituce pohlavních chromosomů u samic motýlů pomocí kombinace genomové in situ hybridizace (GISH) a fluorescenční in situ hybridizace s telomerickou sondou (TTAGG)n (telomerická FISH). GISH u pachytenníchpohlavních chromosomů identifikovala heterochromatin chromosomu W a telomerická FISH detekovala konce chromosomů. Touto metodou jsme ukázali, že Antheraea yamamai má pár pohlavních chromosomů W-Z. U Orgyia antiqua jsme potvrdili přítomnost chromosomů neo-W a neo-Z. Samice Orgyia thyellina vykazovaly konstituci pohlavních chromosomů neo-ZW1W2. U samic Samia cynthia subsp. indet. byl zjištěn trivalent neo-WZ1Z2. Kdežto samice S. c. walkeri měly bivalent neo-W/neo-Z a samice S. c. ricini univalent chromosomu Z (systém Z/ZZ). Kombinace GISH a telomerické FISH nám umožnila nejen získat věrohodné informace o konstituci pohlavních chromosomů, ale i proniknout do evoluce pohlavních chromosomů motýlů.
