Acclimatization of the somatic hybrids in the Solanum genus
Original language description
Many valuable resistance genes are carried by wild species of Solanum genus that are often non-crossable with cultivated potato. Somatic hybridization is an approach overcoming this problem. At our workplace we developed many somatic hybrids using of protoplast fusion by an electric field (Navrátilová B., Greplová M. et al. 2006). Somatic hybrids were evaluated by flow cytometry, RAPD technique and their in vitro morphology was assessed. For further assessment of phenotype an evaluation should be done under in vivo conditions. Optimal method for the most effective transfer into in vivo conditions was investigated. A set of somatic hybrids of two combinations was planted into seedling pots (3x3 cm) containing sterilized soil and cultured in a laboratorywith artificial illumination (4000 Lux) of 16 h light/8 h dark photoperiod at 21 °C. The second variant involved direct planting into the pots (8x8 cm). Plants planted in this way were partially protected by non-woven fleece and exposed
