Original language description

The aim of this work was to induce somatic embryogenesis from leaf segments of Dendranthema grandiflorum - hybrid for transformation by Agrobacterium tumefaciens. We studied optimum conditions of somatic embryogenesis for two clones - 19/93 and 327/95 (pot plants), and one cultivar Reagan (cut plant). The leaf midrib explants from in vitro conditions were cultured on modified MS medium (Murashige and Skoog, 1962). It the experiments the influence of liquid and solidified media, sucrose concentration, phytohormones, effect of light and darkness were tested. Somatic embryos were obtained in the clone 19/93 and in the cultivar Reagan on the solidified medium at the high concentration of sucrose.
