Apoptosis in HL-60 human leukaemia cells treated in vitro with idarubicin (4 hours incubation) was quantified using morphologic changes in the nuclear and cytoplasmic structures in Diff-Quik stained cytospin preparations, flow cytometric measurement of sub-diploid DNA content (sub-G1 cells) and increase of staining with APO 2.7 antibody in nonpermeabilised cells. After treatment by high (20-100 nmol) doses of idarubicin HL-60 cells were found to die earlier, primarily by apoptosis, regardless of the cell-cycle phase, in which they were affected. We have observed that after 6 hours of incubation with small (10 nmol/l) idarubicin dose most of the live cells were in S phase of cell cycle (62%), after 24 hour most of them moved to G2 phase (61%). Increaseof apoptotic cells (20%) was observed as late as 48 and 72 hours after 4 hours incubation with idarubicin. After 72 hours 50% of cells were necrotic (Trypan blue staining).
