Toward the construction of Francisella tularensis 2-DE database
Original language description
Francisella tularensis is an obligate intracellular pathogen whose genome sequencing will be completed shortly. The next step should be the analysis of functional complement of genetic information so called proteome. Proteom is a new rapidly emerging field of study utilizing the combination of two-dimensional gel electrophoresis (2-DE) and mass spectrometry. We applied 2-DE methodology with immobilized pH gradients in the first dimension for separation of complete protein lysates of attenuated Francisella tularensis strain LVS. Two different techniques 9-16 % gradient SDS-PAGE and Tricine SDS-PAGE were used in the second dimension for the resolution of proteins in the range of molecular weights 10 to 200 kDa and 5-70 kDa, respectively. In the first caseabout 2700 proteins were detected on one gel, regarding Tricine SDS-PAGE we were able to stain approximately 1200 different protein molecules. Besides the cultivation of bacteria under normal condition we also examined the protein expres
