Original language description
A set of dominant and co-dominant PCR markers was used to detect the Vf alleles. This methodological procedure was verified on a collection of apple tree varieties with a declared resistance against scab, determined by the dominant Vf gene. Nine reference varieties containing the Vf gene were used, developed by the Research and Breeding Institute of Pomology in Holovousy. Conducted was also a segregation analysis of seedlings of eight complex combination crossbreeds. The resistance or sensitivity of theassessed plant material was also classified in standard greenhouse infection tests. The so-called multi-PCR was used to detect alleles of the Vf gene. The actual amplification and sequences of primer pairs were obtained from Tartarini et al. (1999) exp eriments. The multi-PCR used allowed a simultaneous amplification of one dominant and one co-dominant marker.
