Reprodukce dubu letního (Quercus robur L.) pomocí organogeneze

  • Original language description

We tested a method of in vitro micropropagation of Q. robur L. Primary explants were axillary shoots. Multiple shoots were induced on Woody plant medium (WPM) and MS medium supplemented with 6-benzylaminopurine (BAP 2.25 - 22.5 μM). The highest shoot proliferation rate was achieved on WP medium supplemented with BAP (13.5 μM). Rooting of regenerated shoots involved a two-step procedure. During the first phase microshoots were treated with varying concentration of indole-3-butyric acid (IBA 0-80 μM) for 12 hr, 24 hr and 48 hr. In the second phase microshoots were transferred to a plant growth regulator-free half-strength WP medium. The best results of rooting were achieved when microshoots were treated with 60 μM IBA for 24 hr and then placed on PGR-free half-strength WP medium.
