Development of an enzyme immunoassay for the determination of isoflavones
Original language description
Isoflavones are plant secondary metabolites typical for leguminous plants, however they were found also in certain non-leguminous taxa. Isoflavones were reported to display a wide scale of biological effects in plant-consuming animals including humans. They are endocrinologicaly active due to interaction with estrogen receptors and due to modulation of several hormonogenic metabolic pathways. Genistein is well known inhibitor of tyrosine-protein kinases - the enzymes acting in post-receptor events of peptide hormone signaling. Dietary isoflavones have been reported protective against several types of neoplasias, cardiovascular diseaeses and menopausal symtoms. Enzyme-linked immunosorbent assays (ELISAs) were developed for determination of the most important isoflavones - i.e. genistein, daidzein and biochanin A in biological materials. The indirect competitive ELISA format uses polyclonal rabbit antibodies formed against conjugates of carboxymethyl isoflavones with bovine serum albumin
