    Methods for study of isoflavonoids in non-leguminous plants

    Isoflavonoids (3-phenylchromones) are most abundant in the Fabaceae, however they were recorded in over 20 other families, dicotyledons (e.g. Rosaceae, Asteraceae) as well as monocots (e.g. Iridaceae, Poaceae). Molecular biology data indicate occurrenceof genes for isoflavonoid metabolic pathway even in higher number of families, nevertheless it is difficult to obtain the evidence of their biochemical activity. In our recent work, we combine HPLC-MS and immunochemical approaches. A set of five immunoassays for early products of isoflavonoid metabolic pathway (i.e. daidzein, formononetin, genistein and biochanin A and their glycosides) was adapted for phytochemical studies. Due to the narrow specificities and high sensitivities of these assays (detection limit below 10 pg), the initial screening may be performed in a large number of simply prepared samples, e.g. in diluted water-ethanolic extracts.
