Original language description
This study was focused on a development and an application of a rapid and reliable staining method for characterisation of Saccharomyces cerevisiae cells. Three different fluorescent probes such as propidium iodide (PI), fluorescein diacetate (FDA) and fluorescein isothiocyanate (FITC) were used for staining. PI was used to determine cell viability in a native sample and DNA content in sample fixed by ethanol. To assess protein distribution in yeast population the FITC amine-reactive probe was used. Instantaneous cell enzyme activity was measured as an amount of fluorescein liberated from FDA by intracellular esterase activity.

Czech description
Tato práce byla zaměřena na optimalizaci a aplikaci vhodných metod fluorescenčního značení k charakterizaci buněk Saccharomyces cerevisiae. Využita byla tři různá fluorescenční barviva, propidium jodid, k determinaci buněčné viability a obsahu DNA, fluorescein diacetát k měření okamžité enzymové aktivity a fluorescein isothiokyanát ke sledování obsahu intracelulárních bílkovin.
