In contrast to other retroviruses as HIV, Mason-Pfizer monkey virus (M-PMV) preassembles immature capsids in the cytoplasm. We have performed the in vitro assembly of purified bacterially expressed proteins to address the difference of morphogenically distinct retroviruses M-PMV and HIV in their ability to assemble VLPs from minimized assembly competent domains. The fusion capsid and nucleocapsid protein (CANC) and its N-terminally modified mutant ( ProCANC) were used to mimic the assembly of mature andimmature particles, respectively. We found dramatically different ability to assemble CANC particles between these two viruses. Also ProCANC particles of M-PMV differed from that of HIV forming spheres and tubes, respectively. The requirements for factors affecting assembly were also evaluated and the amount of incorporated fluorescently labeled nucleic acids was determined.
