    Characterization of bacteria in real soil contaminated with persistent halogenated compounds

  • Original language description

    Phytoremediation and rhizoremediation have appeared to be efficient ways of removing persistent halogenated compounds, e.g. polychlorinated biphenyls (PCBs), from soil. In our study, we characterized bacteria in a horse radish (Armoracia rusticana) rootzone contaminated with PCBs. Possible PCB-degrading strains were selected by cultivating on mineral medium with biphenyl. According to 16S rRNA gene sequence analysis Arthrobacter sp., Achromobacter sp., Serratia sp. or Sinorhizobium sp. were identified.Further characterization comprised screening for the presence of bphA1 gene (which encodes the first enzyme of the bph operon) and ribosomal protein profiling using MALDI-TOF MS. This method allows much easier, faster and cheaper identification of bacteria, which can be differentiated at the genus, species, and strain level.

    Fytoremediace a rhizoremediace jsou efektivní způsoby odstraňování perzistentních halogenovaných sloučenin, např. polychlorovaných bifenylů (PCB), z půdy. V naší studii jsme charakterizovali bacterie v kořenové zóně křenu selského. Potenciální degradéřiPCB byly selektovány růstem na minimálním médiu s bifenylem. Sekvenací genů kódujících 16S rRNA byly identifikovány Arthrobacter sp., Achromobacter sp., Serratia sp. nebo Sinorhizobium sp. Další charakterizace zahrnovala screening na přítomnost bphA1 genu a profilaci ribosomálních proteinů pomocí MALDI-TOF.
