Determination of post-mortem age from human tissue in forensic medicine
Original language description
The estimation of the age of an individual is an important task when identifying a corpse of unknown identity. The methods applied can be divided into morphological and biochemical ones. We used biochemical methods for estimation of human age based on determination of posttranslational modification of collagen and noncollagen proteins. Some of the posttranslational modifications cumulate in the aging organism and can be therefore used to determine the age. For determination of age we used: Changes of colour of different human tissue. Determination of pentosidin (one advanced glycation end products). Determination of ratio of the d- and l-forms of aspartic acid by peptide mapping and by HPLC methods. We developed new method based on peptide mapping of noncollagen proteins. Studied tissue: teeth, intervertebral disc Achilles tendon and rib cartilage. The new method applies staphylococcal protease V8, which specifically splits peptide bonds in which l-glutamic or l-aspartic acid partici
