    Preparation of galacto-oligosaccharides using membrane reactor

  • Original language description

    Galacto-oligosaccharides have many biological functions, e.g. selective improvement of growth of beneficial bifidobacteria, improved absorption of calcium and magnesium, and improved detoxification of the body. Galacto - oligosaccharides are produced from lactose by a glycosyl transfer of one or more D-galactosyl units onto the D-galactose moiety of lactose catalysed by ?-galactosidase. Many factors have influence on this reaction e.g. temperature, origin of enzyme, concentration of substrate. This reaction can be carried out in batch and continuous system. We used membrane reactor with ultrafiltration ceramic membrane (150 KDa). Lactose in phosphate buffer (200 g.L-1), recombined whey (197 g.L-1) and ultrafiltration permeate (320 g.L-1) were used as substrate. Concentration of used enzyme Maxilact LX 5000 was 2 g.L-1.     Galaktooligosacharidy mají četné biologické funkce, např. selektiní zlepšení růstu bifidobakterií, zlepšenou absorpci vápníku a hořčíku. Galaktooligosacharidy jsou produkovány z laktosy přenosem galaktosy, který je katalyzován ?-galakosidasou. Reakce může probíhat vsádkovým nebo kontinuálním způsobem. Jakosubstrát byla použita laktosa v pufru, obnovená syrovátka a zahuštěný ultrafiltrační permeát. Nejvyšší koncentrace galaktooligosacharidů byla získána v ultrafiltračním permeátu (41,2 g.L-1 vsádkovým způsobem, 27 g.L-1 kontinuálně).
