Original language description
The aim of this study is novel isolation, purification and structural characterisation of polysaccharides from Pleurotus spopo. The mushroom powder was washed with organic solvents (ethanol, acetone) followed by hot/cold water extraction. Then another extraction of polysaccharides with 1 mol l(-1) aq. NaOH was carried out. The alkali extracts were merged and dissolved in DMSO. The cold water extract was used for purification on DEAE column and then separated according its molecular weight on GPC. The first fraction was determined as a mixture of alpha and beta glucans, the second one mostly contained mannogalactans. The DMSO soluble fraction contained (1 -> 3)-alpha-glucan as the main fraction. The insoluble fraction after all extractions was identified as a chitin-glucan complex. Selected fractions were characterised by FTIR and NMR spectroscopy.
