DEVELOPMENT OF AN EXPERIMENTAL MICRODEVICE FOR DETECTION OF IGG
Original language description
Microfluidic devices are becoming an important tool in diagnostics of diseases and analysis of biological liquids. In this work, we present our progress in development of a microchip for ultra-fast and quantitative determination of human antibodies (IgG)in an aqueous sample. The method is based on the specific biochemical interaction between IgG and the immobilized antigen (Protein A). The antibody molecules are fluorescently labeled and thus their concentration in the microdevice can be monitored by fluorescence microscope. The microdevice consists of an active polystyrene part (PS) with a thin microcappilary and a part for connection of the PS part to the sample and the washing solutions. The microcapillary has diameter between 10 and 20 microns. Use of the electrokinetic phenomena (electroosmosis) for transportation of liquids in the PS part is intensively studied. All step of the immunoassay are described and the experimental determination of IgG concentration in a sample by means
