Diplacone (DPL) is natural flavanone with potential antioxidant, antiphlogistic and antibacterial activity. These beneficial properties can be use in treatment inflammatory diseases, e.g. inflammatory bowel diseases. However, the diplacone is poorly soluble in water which means it has low bioavailability in an organism. Therefore, the delivery of diplacone is challenging. In this work, we propose the use of glucan particles (GPs) as promising carriers for diplacone. Glucan particles are obtained from Saccharomyces cerevisiae (baker’s yeast); they are mainly composed of homopolysaccharides (>85% β-glucans) and they exhibit immunomodulatory properties. The encapsulated drug into GPs can potentially be distributed through the lymphatic system into inflammatory tissue and decrease inflammation due to its activity. The porous GPs, containing diplacone as an active substance, can deliver their content into the targeted inflammatory area and increase its bioavailability. The loaded microparticles were prepared by mixing a solution of diplacone with GPs and subsequently removing the solvent by controlled evaporation with demanded loading efficiency. The DPL/GPs composites were characterized by optical microscopy, SEM, XRD and encapsulation efficiency. The cytotoxicity and anti-inflammatory activity were tested in vitro on macrophage-like cells. GPs with incorporated diplacone provide a promising delivery system, especially for diseases connected with the immune system, and can be used in the treatment of inflammatory diseases in the future.
