The interleaflet transport of phospholipids is a crucial physiological process in the cell plasma membrane. This process can be facilitated by specific proteins—scramblases—without ATP consumption. The experimental data provided here describe the lipid scrambling activity of the Sec61/TRAP translocon, a protein complex previously known for facilitating the membrane translocation of nascent polypeptide chains. In this study, we reconstituted Sec61/TRAP into large unilamellar vesicles and employed two complementary fluorescence-based assays—NBD fluorescence quenching by bovine serum albumin and sodium dithionite—to investigate its scrambling activity. Both approaches confirmed that Sec61/TRAP facilitates lipid scrambling. Notably, blocking the Sec61 translocation cavity did not inhibit scrambling, suggesting the presence of an alternative lipid transport pathway within the translocon complex.
