Original language name
Enzymatic polymerization of phosphonate nucleosides
Original language description
A possibility of enzymatic polymerization of 5?-O-phosphonomethyl-2?deoxynucleosides (PMdN) was studied. Thymine and uracil analogues (PMdT, PMdU) could only be polymerized enzymatically to a limited extent. Adenine and cytosine analogues (PMdA, PMdC) could be incorporated into a DNA duplex with complete chain elongation by all DNA polymerases tested. Vent(exo-) and Therminator polymerases were used for the polymerization.

Enzymatická polymerizace fosfonátových nukleosidů
Czech description
Byla studována možnost enzymatické polymerizace 5´-O-fosfonomethyl-2´-deoxynukleosidů (PMdN). Analoga thyminu a uracilu (PMdT, PMdU) byla enzymaticky polymerizována pouze v omezeném rozsahu. Analoga adeninu a cytosinu (PMdA, PMdC) byla inkorporována do DNA duplexu s kompletním prodloužením řetězce všemi DNA polymerasami. K polymerizaci byly použity Vent(exo-)- a Therminatorpolymerasa.
