Original language description
Protein three-dimensional structure determination by X-ray crystallography is important for understanding protein function. The protein crystallization bottleneck can be overcome by optimization of factors which influence the protein crystallizability. We demonstrated this approach on two scFv antibody fragments, and on prokaryotic metabolic regulator. We have performed protein pre-crystallization analysis employing the combination of size-exclusion chromatography, dynamic light scattering, and thermofluor assay. Based on the analysis data, we have developed crystallization protocols which yielded monocrystals suitable for X-ray diffraction analysis. This enabled us to determine crystal structure of the effector-binding domain of DeoR repressor.
