    Yersinia outer protein P (YopP) of Yersinia enterocolitica induces apoptotic and necrosis-like cell death in dendritic cells (DC). YopP induced the cleavage of pro-caspase 8 and c-FLIPL degradation independently of tumor necrosis factor (TNF) receptor family members since the expression of both CD95/Fas/APO-1 and TRAIL-R2 on DC was low. Moreover, DC from TNF-Rp55-/- mice were not protected against YopP-induced cell death. By coimmunoprecipitation we found that both cleaved caspase 8 and receptor interacting proteins 1 (RIP1) were associated with the Fas-associated death domain (FADD) indicating the formation of an atypical death-inducing signaling complex (DISC). Altogether our data suggest that Yersinia-induced DC cell death is independent of death domain cointaing receptors but mediated by RIP1 and caspase 8 at the level of DISC

    Patogenní protein YopP (Yersinia outer protein ? Yop) bakterie Yersinia enterocolitica způsobuje apoptózu a nekróze podobnou buněčnou smrt u dendritických buněk (DC). YopP spouští štěpení kaspázy 8 a c-FLIPL proteinu nezávisle na receptorech z rodiny tumor nekrotizujících faktorů (TNF). Bylo zjištěno, že DC exprimují na svém povrchu velmi málo receptorů CD95/Fas(APO-1 a TRAIL-R2. Navíc, DC izolované z TNF-Rp55-/- myši byly stejně náchylné k buněčné smrti způsobené YopP. Pomocí koimunoprecipitace bylo zjištěno, že rozštěpená kaspáza 8 i receptor interagující protein 1 (RIP1) interagují s FADD proteinem, čímž dochází k vytvoření atypického signalizačního komplexu indukujícího buněčnou smrt (DISC). Tyto data ukazují, že buněčná smrt u dendritických buněkvyvolaná bakterií Y. enterocolitica nezávisí na signalizaci přes receptory indukujících buněčnou smrt, ale je signalizována pomocí proteinu RIP1 a kaspázy 8 na úrovni DISC
