Bioactive support for cell cultivation and potential grafting. Part1. Surface modification of 2-hydroxyethyl methacrylate hydrogels for avidin immobilization

  • Original language description

Synthetic hydrogels are often used in biomedical applications as many of them are compatible with living tissue and moreover they meet the most criteria for artificial tissue properties. To enhance the cell adhesion and proliferation, it is desirable tomodify the polymer surface by some bioactive compounds (e.g. saccharides, proteins). In this work, firstly carboxyl-enriched hydrogels were prepared by various methods: by copolymerization of 2 hydroxyethyl methacrylate with methacrylic acid, by hydrolysis or oxidation of poly(2 hydroxyethyl methacrylate). Subsequently, avidin was immobilized to the modified hydrogel surfaces either electrostatically through dissociated carboxylic groups or covalently through carboxylic groups activated by N-hydroxysuccinimide. Preliminary biological tests showed that hydrogels with immobilized avidin provides better adhesion and proliferation of keratinocytes compared to supports without avidin.
