Original language description

The benefits of the use of floating membrane rafts (Osmotek, Israel) for the cultivation of embryogenic cultures of Norway spruce were shown. The system allows perfect nutrient uptake in a similar way as in suspension culture, the labor costs are reduced. On the other hand, the system holds the advantages of the systems using solidified media (better air exchange, stable orientation of the culture). On the floating membrane rafts, the development of majority of cell lines was accelerated and the yield of mature somatic embryos was increased compared both to the cultivation on solidified or liquid media. Subsequent germination of somatic embryos was not affected.

Czech description

Bylo prokázáno, že pro kultivaci embryogenních kultur smrku ztepilého (Picea abies) lze s výhodou užít systém tzv. plovoucích membránových prámků, membrane rafts (Osmotek, Izrael). Tento způsob kultivace kombinuje výhody systému pěstování v suspensi (tj.dokonalý přísun živin, snadnější manipulace) s výhodami systému kultivace na pevných médiích (zejména lepší přístup vzduchu, stálá orientace kultury). Kultivací na plovoucích membránových prámcích bylo u většiny buněčných linií dosaženo zrychlení vývojekultury a zvýšení výtěžku zralých somatických embryí, aniž by byla snížena jejich klíčivost.
