    Occurrence of Phytoplasmas in Red and White Currant in the Czech Republic

    The presence of phytoplasmas in the shrubs of red ('Vitan', 'Losan', 'Mulka', 'Rubigo', and some of new selections) and white ('Primus') currant exhibiting full blossom symptoms was estimated using nested PCR with primers amplifying phytoplasma 16S rRNAgene and by electron microscopy. In total, 20 of 41 analysed symptomatic bushes tested positive phytoplasma in PCR. Based on the results of RFLP analysis and sequencing the phytoplasma isolates were identified as belonging to the apple proliferation andaster yellows groups. European stone fruit yellows phytoplasma was the most common phytoplasma identified in currants in this study. Furthermore, apple proliferation phytoplasma, aster yellows phytoplasma and pear decline phytoplasma were detected. The presence of phytoplasma bodies in phloem tissue of red currants was confirmed by electron microscopy.

    Pomocí PCR a elektronové mikroskopie byla sledována přítomnost fytoplazem v kultivarech červeného a bílého rybízu, vykazujících plnokvětost. 20 ze 41 testovaných keřů bylo v PCR pozitivních. Detekované fytoplazmy byly na základě RFLP a sekvenování zařazeny do skupin fytoplazmy proliferace jabloní a fytoplazmy žloutenky aster. ESFY byla nejběžnější fytoplasmou identifikovanou ve vzorcích rybízu., nicméně byly detekovány I fytoplazma proliferace jabloní, f. žloutenky aster a f. chřadnutí hrušní.
