    Present Status of Research on the Etiology of the Full Blossom Disease in Red and White Currants in the Czech Republic

    The presence of phytoplasmas in the shrubs of red and white currants exhibiting full blossom symptoms was determined by nested PCR with primers amplifying phytoplasma 16S rRNA gene, sequencing and by electron microscopy. In total, 6 of 10 analysed symptomatic bushes gave positive PCR reaction. Based on the results of sequencing the phytoplasma isolates were identified as belonging to the apple proliferation group and aster yellows group. The presence of phytoplasma-like bodies in the phloem of two red currant bushes infected with aster yellows and apple proliferation phytoplasmas was confirmed by electron microscopy. All 10 analysed bushes were to be found infected with the Blackcurrant reversion virus. In graft experiments, the disease was transmittedfrom infected bushes to red currant ´Rondom´, ´Jonkheer van Tets´ and ´Vitan´, white currant ´Blanka´ and black currant ´Viola´.     Pomocí PCR, sekvenování a elektronové mikroskopie byla sledována přítomnost fytoplazem v keřích červeného a bílého rybízu vykazujících plnokvětost. Na základě sekvenování byly detekováno fytoplazmy určeny jako fytoplazma žloutenky aster a fytoplazma proliferace jabloní. Tělískap odobná fytoplazmám byla identifikována I pomocí elektronové mikroskopie. U všech deset testovaných keřů byla detekována přítomnost viru zvratu rybízu. V roubovacích experimentech byl potvrzen přenos choroby na zdravé rostliny.
