The aim of this work was to examine the effect of accession or genotype, source of protoplasts and culture technique on protoplast culture. Protoplasts of selected genotypes of Cucumis and Cucurbita were isolated and cultured. As sources of protoplasts were used young leaves, hypocotyls, etc. Various cultivation media were tested.

Czech description
Cílem práce bylo testování vlivu genotypu, donorové části rostliny a složení médií na izolaci a kultivaci protoplastů dvou rodů čeledi Cucurbitaceae - Cucumis a Cucurbita. Byly testovány vybrané genotypy obou druhů; jako donorové části byly použity mladélisty, hypokotyly, kalusy, atd; dále byly testovány vybrané typy médií doplněných o růstové regulátory. Byla vyhodnocena úspěšnost izolace a kultivace protoplastů a porovánány výsledky u jednotlivých genotypů.
