Comparison of two embryogenic cell lines of silver fir (Abies alba Mill.)
Original language description
The embryogenic cell lines L27 and L122 were initiated from different immature zygotic embryos of silver fir (Abies alba Mill.). No polymorphism was found between these two cell lines by DNA analysis, but embryogenic cell lines differed during proliferation and maturation process. Embryogenic tissue L27 has more quickly turned brown and necrotized after 27 days of proliferation. These cell lines differed also in number of mature somatic embryos obtained by maturation process. Production of cotyledonarysomatic embryos and regenerants was significantly higher in L122 cell line. Protein content was higher in cell line L122. It was confirmed that inhibitory substances from pre-cultivated somatic embryos L27 spreading to agar cultivation medium by simple diffusion cause greater inhibition of tester mycelial growth during subsequent co-cultivation, compared to somatic embryos L122.
