The process o fleukocyte cytolysis and the manifestations of apoptosis and secondary necrosis of neutrophil granulocytes (hereafter only "neutrophils") were studied on four virgin heifers after the induction of leukocyte influx into the mammary glandandafter their lavage in in vitro conditions. Phosphate buffered saline,muramyl dipeptide and a lipopolysaccharide were used for influx induction.Cytolys is and apoptosis were induced with heat stress, ultraviolet irradiation and spontaneous aging for 24 hours. The cytolysis was detected indirectly by determining the lactate dehydrogenase activity in the cultivation medium after the enzyme was released through cell lysis. The neutrophil apoptosis was detected using flowcytometry and two staining methods(i)simultaneous staining with Annexin V labelled FITC and propidium iodide and (ii) with SYTO 13. Proces cytolýzy leukocytů a projevy apoptózy a sekundární nekrózy neutrofilních neutrofilních granulocytů (dále neutrofilů) byly studovány s použitím čtyř virginních jalovic po indukci influxu leukocytů do mléčné žlázy a jejich laváži v in vitro podmínkách. K indukci influxu byly použity phosphate beffered saline, muramyl dipeptide a lipopolysacharid. Cytolýza a apoptóza byly indukovány tepelnou zátěží, ultrafialovým ozářením a spontanním stárnutím po dobu 24 h. Cytolýza byla detekována nepřímo stanovením aktivity laktat dehydrogenasy v kultivačním mediu po uvolnění enzymu lýzou buněk. Apoptóza neutrofilů byla detekována průtokovou cytometrií dvěma postupy barvení i) souběžným obarvení FITC značený Annerinem V a propidium jodidem a ii) SYTO 13. Bylo zjištěno, že leukocyty mléčné žlázy virginních jalovic podléhají během inkubace in vitro cytolýze spontánním stárnutím.
