The porcine zona pellucida is composed of three glycoproteins called ZP2, ZP3 and ZP4, and the presence of carbohydrate moiety is crucial for sperm-egg recognition. After fertilization, the zona glycoproteins are proteolytically cleaved and occurs so-called zona hardening to prevent polyspermy. But during in vitro fertilization of porcine oocytes matured in vitro this process fails. The ZP2 and ZP3 protein was visualised using selected antibodies ZP2 M-19 and ZP3 N-20. The antibody using ZP2 emerged asnonspecific. However, efficacy of the stripping strategy using mercaptoethanol was demonstrated. Ideal temperature of stripping for zona proteins was determined to be 50oC. Then there is a loss of target protein.
