Original language description
Flow cytometric analysis of suspension of isolated chromosomes enables classification of chromosmes in the form of histograms (called flow-karyotypes) based on biochemical features such as their relative DNA content, base content, protein content, and various morphological parameters. Flow karyotyping has found applications in monitoring karyotype changes, for the detection of aberrant chromosomes, and for mutagenicity testing. A high-yield method for isolation of Norway spruce chromosomes in suspension, their analysis and sorting using flow cytometry is described. To accumulate meristem root tips cells at metaphase, actively growing roots were subjected treatments with 0,05 % colchicine for 24h (Überall et al. 1997).
