    A new procedure with supercritical CO2 modified with 0.5 ml water and 0.75 ml 0.1 M HCl in situ and 0.75 ml of water on-line (7.5 % v/v) at 15 MPa, 50 °C, for 45 min was applied for extraction of bio-available amino acids from soil samples. Total extraction time was 60 min but more favourable conditions are even possible for selected groups of amino acids. All analytes were trapped into 20 ml of MeOH with satisfying recovery (94 % - 104 %) and determined using high-performance liquid chromatography withfluorimetric detection (HPLC/FLD) on Zorbax Eclipse column (4.6 x 75 mm, 3.5 micrometers) with Na2HPO4 and ACN:MeOH:water as a mobile phase. Linear calibration curves were obtained (r is higher than 0.999 except 0.99823 for Ile) with lower limits of detection in the range of 1.54 pg (Gly) - 13.45 pg (Cy2), or 18.6 fmol (Ser) - 64.8 fmol (Lys).     Nový postup se superkritickým CO2 modifikovaným 0,5 ml vody a 0,75 ml 0,1 M HCl in situ a 0,75 ml vody on-line při 15 MPa a 50 C po dobu 45 min. byl použit pro extrakci biologicky přístupných aminokyselin z půdy. Celkový čas extrakce je 60 min., příznivější podmínky jsou možné pro vybrané skupiny aminokyselin. Všechny analyty byly zachytávány do 20 ml methanolu, s dostatečnou návratností (94 - 104 %) a stanoveny HPLC chromatografií s flourescenční detekcí (Zorbax Eclipse column (4.6 x 75 mm, 3.5 micrometers) with Na2HPO4 and ACN:MeOH:water as a mobile phase). Lineární kalibrační křivky byly získány (r je vyšší než 0.999 kromě 0.99823 v případě Ile)s nižšími limity detekce (S/N=3)v rozsahu od 1.54 (Gly)do 13.5 pg (Cy2)nebo od 18.6 fmol (Ser) do 64.8 (Lys). Srovnatolné výsledky byly dosařeny aplikací relativně robustní metody (0,5 m octan amonný, 60 min. třepání, filtrace a lyofilizace).Byly stanoveny limitované hodnoty kontaminace aminokyselinami, které nepocházely z půdního zdroje.
