    SSR analyses within Prunus genus

  • Original language description

SSR polymorphisms are extensively used as genetic markers. They are generally codominant, highly polymorphic, they have high eficiency in a species with low variability such as peach and can be detected with a simple PCR procedure. Consequently SSR wereused for study of the genetic diversity in Prunus genus. The 9 known SSR pair of primers nominated by other autors were applied to 17 Prunus cultivars. SSR analyses were performed with the thermal cycler program by the use of touchdown protocol for annealing The electrophoretic spectrum of the obtained bands on the agarose MetaPhor were similar to the spectrum on polyacrylamide (PAGE). The gel on the Metaphor was visualised by ethidium bromide staining, the polyacrylamide gel by the SYBR Gold. The primers distinguished few a homozygous and heterozygous genotypes in given locus within all analysed cultivars. The given sizes of the SSR products were compared with the resulting product sizes in work of Testolin et al. (2000). These results
