Original language description
The anticoagulant effect of heparin is expressed through activation of antithrombin (AT). We have developed a novel covalent AT-heparin complex (ATH) that has higher specific activity than unfractionated heparin (UFH) (750 and 180 U/mg, respectively) andinhibits thrombin more rapidly than equimolar concentrations of AT plus UFH (second order rate constants of 3.1 x 109M-1s-1 and 1.0 x 108M-1s-1, respectively). Because of its superior anticoagulant properties, ATH may be better than UFH for prevention ofarterial thrombosis. To explore this possibility, we compared the efficacy and safety of ATH with UFH in rabbit arterial thrombosis prevention and bleeding ear models.
