Hoof diseases in dairy cows are an important research topic, where the most common problem is laminitis. Complete causes of this disease are still not fully understood. The critical period for the formation of laminitis is after the parturition when is increased demands for milk production and the food composition changes. This can cause a rumen acidosis where the rumen microflora also produces histamine, acting mainly on local microcirculation in hoofs. Many studies in vivo and in vitro have shown thatkeratinocytes are capable to produce matrix metalloproteinase -2 (MMP -2), which expression during laminitis is increased, and the cytokine IL- 1, which triggers a cascade of inflammation retroactively. Purpose of the project was induction cultures of bovine keratinocytes by various concentrations of histamine.
