The effect of azotemia on erythrocyte osmotic fragility in dogs with chronic and end-stage renal failure.

Objective: Dogs with chronic renal failure (CRF) may suffer from extreme azotemia. Urea enters erythrocytes (RBC) through its membrane. Once there, it destabilize proteins and change the set point of volume of RBC to lower values. The RBC with smaller MCV is more resistant to hypotonic environment. Previous studies on osmotic fragility revealed contrarily results. The aim of this study was to find an effect of azotaemia caused by chronic renal failure on RBC in dogs. Methods: Thirteen dogs with CRF andeighteen healthy dogs were enrolled in this study. Twenty-five microliters of heparin blood was mixed with 2,5ml of NaCl solution with decreasing concentration 0,9% - 0,1%. After incubation of 30 minutes and centrifugation the absorbance of the supernatant was measured by ELISA reader at wavelength of 540 nm. The cumulative curve was made.
