    Identification of E. coli resistant to beta-lactam antibiotics isolated from Czech diary calves

    Antimicrobial resistance is a major threat to human and animal health. Our study focused on beta-lactam resistance in Escherichia coli from calves in Czech farms. In 2019, rectal swabs from 266 calves across 24 farms were collected, and resistant E. coli were isolated using cefotaxime-supplemented McConkey agar. Susceptibility to 13 antibiotics was assessed by disc diffusion, and resistance genes were detected by PCR. A total of 128 (48%) E. coli isolates were obtained, with 90% (115/128) being multidrug-resistant. All isolates were resistant to ampicillin, and 127 to cephalotin. Resistance rates to other antimicrobials ranged from 13% (gentamicin) to 73% (tetracycline). ESBL production was confirmed in 91% (117/128) of isolates, while 36% (46/128) produced AmpC. PCR analysis revealed the presence of blaCTX-M (90%), blaTEM (48%), and qnrS (21%) genes. Whole-genome sequencing (WGS) was performed on 68 isolates from 16 farms. Our findings highlight the high prevalence of multidrug-resistant E. coli carrying clinically relevant resistance genes, emphasizing the need for antimicrobial stewardship in animal production.
