The focus of our work is the structure-specific binding of wild-type p53 and mutant p53 to DNA containing quadruplex-forming sequences in vitro and in cells and the effect of drugs on p53-DNA binding and p53 driven transcription [3-6]. Bioinformatics tools was applied for systematic investigation of know wild-type p53 and mutant p53 target gene promoters for G-quadruplex forming regions. Biophysical techniques and DMS footprinting were used for detection of G-quadruplexes in selected sequences and on drug effect on G-quadruplex formation.
