    Three-dimensional Biomaterials as a Tool for In Vitro Culture of Leukemic B Cells

    Chronic lymphocytic leukemia (CLL) is the most common adult leukemia in Caucasians. However, mechanisms underlying the disease development still have not been fully elucidated. To unravel the machinery of neoplastic processes in CLL cells, in vitro culture is inevitable. Construction of a suitable microenvironmentlike in vitro platform therefore represents an important goal in CLL research. An authentic model of CLL can be, for example, achieved by utilization of three-dimensional biomaterials. In our study, we focused on use of porous scaffolds and gelling substances for the growth of cells which are naturally present in blood marrow of CLL patients, i.e., B cells (CLL cells) and bone marrow stromal cells. We observed that P(HEMA-co-AEMA)-RGDS hydrogels, collagen scaffolds, and CMC-PEG gels are biocompatible materials, which enable long-term 3D in vitro culture of HS5, M2-10B4, MEC-1, and HG-3 cell lines and short-term culture of primary CLL cells. While further optimizations of the culture systems are necessary, our methodology and results already represent an important step toward developing a 3D model of CLL.
