    This work comprises characterization of the technique used for rapid cooling and heating of the cells in tissue culture. Principle of the technique is feedback control of temperature of the solution superfusing the cells during the experiment. This system is suitable for the study of temperature dependence of ion channels. The work is accompanied with examples of use of the technique for temperature-activated channels TRPV1, TRPM8 heterologously expressed in HEK cells and sensory neurons in tissue culture

    V této práci je popsána technika pro rychlé chlazení a zahřívání buněk v tkáňové kultuře. Princip tohoto systému spočívá v zpětnovazebné kontrole teploty roztoku omývajícího buňky při experimentu. Tato technika je vhodná pro studium teplotní závislosti iontových kanálů. Práce je doprovázena příklady využití techniky pro teplotně aktivované kanály TRPV1, TRPM8 heterologně exprimované v HEK buňkách a sensorické neurony pěstované v kultuře
