    Advantages and pitfalls of using free-hand sections of frozen needles for three-dimensional analysis of mesophyll by stereology and confocal microscopy

    In our stereological measurements of proportion of intercellular spaces in mesophyll and internal needle surface area by fakir method in stacks of optical sections acquired by a confocal microscope within thick physical free-hand sections, we tested theeffect of freezing conifer needles on their anatomical structure as well as the effect of possible deformations due to the cutting of unembedded material by a hand microtome. We found a higher proportion of intercellular spaces in mesophyll in regions near to the surface of a physical section, which means that the measurements should be restricted only to the middle region of the optical section series. On the other hand, there was no significant difference in mesophyll parameters between fresh and frozen needles, therefore, freezing represents a suitable form of storage of sampled material for proposed stereological evaluation

    Výhody a úskalí při používání ručních řezů zmrazenými jehlicemi pro trojrozměrnou analýzu mezofylu stereologií a konfokální mikroskopií

    Testovali jsme, jaký vliv má zmražení jehlic smrku na jejich anatomickou stavbu a k jak vážným deformacím dochází při krájení řezů jehlicemi ručním mikrotomem, které používáme při stereologickém měření anatomických parametrů mezofylu. Měřili jsme podíl intercelulár v mezofylu a vnitřní povrch jehlice fakírskou metodou, která je aplikována na série optických řezů získaných konfokálním mikroskopem. Zjistili jsme, že ve vrstvách poblíž řezné plochy ručního řezu je podíl intercelulár v mezofylu zvýšený, cožznamená, že měření by mělo být omezeno na řezy uprostřed nasnímané série optických řezů. Na druhou stranu jsme nezjistili žádné změny v anatomických parametrech mezofylu mezi čerstvými a v mrazu uchovávanými jehlicemi, proto lze pro naše měření jehlicetímto způsobem uchovávat
