    Ivermectin, allosteric modulator of ATP-gated PX4 receptor-channels, induces conformational changes of the entire protein that alter potencies and efficacies for orthosteric ligands and transition of ion channel from open to dilated state. The kinetic (Markov state) model indicates that the ivermectin-dependent transition is coupled to receptor sensitization, which rescues the receptor from desensitization and subsequent internalization
