To test the influence of a Nicotiana tomentosiformis repetitive sequence (R8.3) on transgene expression in N. sylvestris and in N. sylvestris-N. tomentosiformis hybrids, the R8.3 sequence was placed upstream of a nopaline synthase promoter (NOSpro)-NPTIIreporter gene in a T-DNA construct. In most transgenic lines the NPTII gene was expressed, however one line (S22dot) showed silencing of NPTII gene expression and methylation of NOSpro sequences. Methlylation of promoter cannot be explained by spreadingfrom adjacent R8.3 region since in silnced S22dot line this region was substantially less methylated than in nonsilenced lines. We conclude that the effect of the dispersed repeat R8.3 family on transgene expression was negligible.
