    Binding of mutant and wild type p53 proteins to supercoiled DNA

    Full length wild-type p53 (wtp53) can interact with DNA via its central (CD) or C-terminal (CTD) domains and it binds selectively to supercoiled DNA (SCS binding) regardless of the presence or absence of the p53 consensus sequence (p53CON). The SCS binding was studied using selective modulation of the CD and CTD by oxidation agents and monoclonal antibodies. Majority of the hot spot mutant p53 proteins (mutp53) lost the SCS binding, but they can still induce or repress transcription of mutp53-specific target genes and they exhibit the ability to interact selectively with non-B DNA elements (e.g., MAR/SAR elements and stem-loop structures).

    Celý standardní protein p53 (wtp53) může interagovat s DNA svou centrální doménou (CD) nebo C-koncovou doménou (CTD) a touto se váže selektivně s superhelikální DNA (SCS vazba). Tato superhelikálně selektivní vazba byla studována pomocí protilátek. Většina nejčastěji se vyskytujících mutantních proteinů p53 ztrácí schopnost sekvenčně specifické vazby, ale stále je schopna aktivovat či potlačovat cílové geny (vlastní mutantním proteinům) a dále jsou mutantní proteiny schopny interagovat selektivně s non-B DNA elementy (např. MAR/SAR oblasti a stem-loop struktury).
