    Recognition of cisplatin-damaged DNA by p53 protein: Critical role of the p53 C-terminal domain

    Protein p53 can recognize DNA modified with cisplatin. We studied p53 binding to cisPt-DNA using p53 deletion mutants and via modulation of binding by changes of protein redox state. C-terminal domain (CTD) bound to the cisPt-DNA with higher affinity than to unmodified DNA. Constructs lacking C-terminal DNA binding site (CTDBS) exhibited small preference for cisPt-DNA. Oxidation of full length p53 did not affect its ability to recognize cisPt-DNA. Blocking of CTDBS by monoclonal antibody resulted in abolishement of the CTD binding to the cisPt-DNA. Our results demonstrate a crucial role of the basic region of the p53 CTD in the cisPt-DNA recognition.

    Protein p53 může rozpoznat DNA modifikovanou cisplatinou. Studovali jsme vazbu p53 na cisPt-DNA s využitím p53 delečních mutantů a modulací vazby změnou redox stavu proteinu. C-koncová doména (CTD) se vázala na cisPt-DNA s vyšší afinitou než k nemodifikované DNA. Konstrukty bez C-koncového DNA vazebného místa (CTDBS) vykazovaly malou preferenci k cisPt-DNA. Oxidace p53 o plné délce neovlivnila jeho schopnost rozpoznávat cisPt-DNA. Zablokování CTDBS monoklonální protilátkou vyústilo ve zrušení vazby CTDna cisPt-DNA. Naše výsledky demonstrují rozhodující úlohu bazické oblasti CTD proteinu p53 v rozpoznání cisPt-DNA.
